评价 Bemarituzumab (FPA144) is first-in-class anti-fibroblast growth factor receptor 2b (FGFR2b) chemotherapy antibody from Five Prime Therapeutics . Amgen acquired Five Prime in 2021. Bemarituzumab is a humanized immunoglobulin G1 (IgG1) isotype antibody. It binds to FGFR2b and promotes ADCC via enhanced binding to FcγRIIIa. Around 30% of HER2-negative gastric cancers are FGFR2b positive and bemarituzumab is designed to target this cancer subtype that uses the FGF/FGFR pathway to promote transformation, proliferation and angiogenesis.

临床描述 This is a global, randomized, double-blind, controlled study to evaluate the efficacy of bemarituzumab (FPA144) + mFOLFOX6 versus placebo + mFOLFOX6 in patients with FGFR2 selected Gastric Cancer (as determined by prospective IHC FGFR2b overexpression and/or a ctDNA blood assay demonstrating FGFR2 gene amplification)
This is a three-part, open-label, safety, tolerability, and PK study of FPA144. Patients will be enrolled in Part 1 (A or B, dose escalation) or Part 2 (dose expansion) of the study, but not both.
This is a multi-center study to evaluate the safety, tolerability, pharmacokinetics (PK), and pharmacodynamics (PD) of FPA144 in combination with 5-fluorouracil, leucovorin and oxaliplatin (mFOLFOX6) for patients with gastrointestinal malignancies (GI). The phase 1 open label safety run-in will identify a recommended dose of FPA144 to use in the phase 3 portion of the trial.
