Double stranded DNA often moves faster in TAE than in other buffers, but it will also be unable to swim due to the depletion of buffer ions during long-term electrophoresis. Buffer circulation or buffer replacement during long-term electrophoresis can compensate for lower buffer capacity. Dilute the concentrated TAE buffer solution to obtain 1 ×? TAE buffer, containing 40mM Tris acetate and 1mM EDTA, pH 8.3. one ×? Tae buffer can be used for both agarose gel and electrophoresis buffer.
